The science behind NAD+ and NMN.
NMN stands for Nicotinamide Mononucleotide — a naturally occurring molecule found in small amounts in certain foods. It is the most direct precursor to NAD+ (Nicotinamide Adenine Dinucleotide), converting to NAD+ in a single enzymatic step inside your cells.
NAD+ is the coenzyme your mitochondria use to convert food into cellular energy (ATP). Without adequate NAD+, cellular energy production, DNA repair, and sirtuin activation all suffer. NMN supplementation is the most studied and direct way to restore declining NAD+ levels.
NAD+ declines for two main reasons: your body produces less of it as you age, and cellular demand for NAD+ increases as DNA damage accumulates. The combination is significant — by age 60, most people have roughly half the NAD+ they had at 20. By 80, less than a quarter.
This decline tracks almost every hallmark of biological aging: mitochondrial dysfunction, reduced cellular energy, suppressed sirtuin activity, accumulating senescent cells, and declining cognitive and physical performance. NAD+ restoration via NMN is one of the most-studied approaches to addressing this at a cellular level.
NMN (Nicotinamide Mononucleotide) and NR (Nicotinamide Riboside) are both NAD+ precursors, but NMN is one step closer to NAD+ in the conversion pathway. NMN converts to NAD+ via the NMNAT enzymes in one step; NR first converts to NMN, then to NAD+.
The human evidence base for NMN is growing rapidly — multiple peer-reviewed trials have demonstrated NAD+ increases in humans taking NMN. The research on NR is more established but covers a different pathway. Many researchers and longevity practitioners — including David Sinclair at Harvard — have shifted to NMN supplementation based on the emerging evidence.
Honest answer: NMN reliably and measurably raises NAD+ levels in humans. This is documented across multiple peer-reviewed human trials. The downstream benefits — improved energy, cognitive function, sleep quality, and longevity — have strong mechanistic rationale and compelling emerging human evidence, but are not yet as definitively established as a drug with 30 years of randomised controlled trials.
We are not selling you certainty. We are selling you the most credible bet in cellular health right now, backed by Harvard research and published human trials. We believe that is worth being honest about — because overclaiming destroys trust, and trust is the only foundation a supplement brand should stand on.
This concern comes from a single animal study using UV-irradiated mice, which found a possible association with melanoma in that specific experimental context. No human evidence of cancer risk from NMN supplementation exists.
In fact, multiple studies suggest NAD+ boosting may enhance immune cell efficacy against cancer. The FDA has confirmed NMN is safe as a dietary supplement at supplementation doses. If you have an active cancer diagnosis or are in active treatment, please consult your oncologist before taking any NAD+ supplement — as you should with any new supplement when managing a serious health condition.
How we verify what is in every capsule.
This is the most common question we receive — and the most misunderstood situation. ConsumerLab independently tested the top 21 NMN brands on Amazon. 64% had less than 1% of their claimed NMN. 14% had zero. Most had thousands of 5-star reviews.
If you took NMN and felt nothing, the most likely explanation is that you were not actually taking NMN. You were taking nicotinamide, or a capsule full of filler, or a product with trace amounts of the real molecule. This is not a failure of NMN as a category — it is a quality crisis in the supplement industry. We publish our Certificate of Analysis so you never have to wonder about ours.
A Certificate of Analysis (CoA) is a document from an independent third-party laboratory confirming the identity, purity, and potency of a supplement batch. It shows what is actually in the capsule — not what the manufacturer claims is there.
At Nadovia, every batch is HPLC-tested (High-Performance Liquid Chromatography — the gold standard for ingredient verification) for NMN purity and potency, β-isomer confirmation, heavy metals (lead, arsenic, cadmium, mercury), and microbial contamination. The CoA is published on our website for every batch and can be downloaded before you buy. We do not ask for your trust — we give you the data.
NMN exists in two structural forms: the β-isomer (the bioactive form used in human trials) and the α-isomer (less bioactive). Some products use racemic NMN — a mixture of both isomers — which effectively dilutes the dose of the bioactive form. Others mislabel plain nicotinamide as NMN entirely.
Nadovia uses exclusively the β-isomer, verified at >99% purity by HPLC. This is what the Harvard and Keio University human trials used. This is the only form we consider acceptable in a clinical-dose formula.
Nadovia is manufactured in a BRC-accredited, GMP-certified facility. GMP (Good Manufacturing Practice) certification requires the facility to meet strict standards for cleanliness, quality control, ingredient verification, and documentation. BRC accreditation is an additional global standard for food and supplement safety.
Every production batch is independently tested by a third-party laboratory — not our own facility, not a related entity. The results are published as a Certificate of Analysis for every batch.
Getting the protocol right from day one.
Most human clinical trials used doses between 250mg and 600mg per day. We formulate at 500mg per serving — the dose with the most robust human evidence for NAD+ elevation. This is one serving of the Longevity Complex (3 capsules) or one capsule of Pure NMN 500mg.
You can increase to 2 Pure NMN capsules (1000mg) after 4 weeks if you wish. At doses above 500mg, we recommend adding TMG (300–600mg) to protect methylation. The Longevity Complex already includes 600mg TMG per serving.
Morning, ideally with your first meal. NAD+ has a circadian rhythm — it is naturally higher in the first half of the day, and NMN supplementation aligns best with this rhythm when taken in the morning.
Food is not required for absorption, but taking NMN with a meal may improve tolerance in those with sensitive stomachs. Do not take it within a few hours of sleep — the energy-supporting effects may interfere with falling asleep for some people.
NMN is generally well-tolerated alongside common supplements including magnesium, omega-3, vitamin D, and collagen. The Longevity Complex pairs well with additional Quercetin or Pterostilbene if you are building a more comprehensive longevity stack.
If you take blood thinners, blood pressure medication, or medication that affects methylation pathways, please consult your healthcare provider before adding NMN to your routine. This is not a standard exclusion — it is standard prudence when adding any new supplement alongside prescription medication.
TMG (Trimethylglycine) is not optional at therapeutic NMN doses. Here is why: every NMN→NAD+ conversion consumes a methyl group from your body's methyl pool. At 500mg NMN per day, this methyl group consumption is significant. Without TMG replenishment, long-term NMN use can deplete methylation capacity — the very biological system that NMN is trying to support.
David Sinclair takes TMG alongside NMN for precisely this reason. We include 600mg per serving — the highest dose we have found in any NMN formula — because it is not a supporting ingredient, it is a non-negotiable part of the protocol.
The honest, clinical timeline.
Most members notice something between weeks two and four. NMN is not a stimulant — it does not produce an immediate, obvious effect on day one. The first signals are typically subtle: slightly less reliance on afternoon coffee, slightly more restorative sleep, marginally faster exercise recovery.
Clearer, undeniable differences — cognitive clarity, sustained energy, exercise performance — typically emerge between weeks four and twelve. Cellular and longevity benefits compound over months and years. We set this expectation upfront because we would rather you understand the timeline than quit at week one and conclude the product does not work.
The most consistent reports from our members, in order of frequency: less severe afternoon energy drop (the 2pm wall softens or disappears), more restorative sleep (falling asleep more naturally, waking feeling more refreshed), faster exercise recovery, and improved cognitive stamina — the ability to sustain focus through long, demanding days.
Importantly, NMN does not feel like caffeine. There is no buzz, no spike, no crash. What most members describe is the absence of fatigue that had become their baseline. The clearest confirmation for most people comes when they forget to take it for a few days and notice the difference.
Yes — and long-term use is the point. All ingredients in Nadovia's formulas have well-established human safety profiles. Multiple human trials have demonstrated NMN's safety at doses up to 1200mg per day with no significant adverse effects. The FDA has confirmed NMN's safety as a dietary supplement.
The TMG inclusion in our Longevity Complex is specifically there to support long-term NMN use by protecting methylation capacity — without it, therapeutic NMN doses can create downstream issues over months of use. We designed this formula for people who intend to take it indefinitely, not just for a trial period.
